tlmgr --help

would give you some indication! You have to change the installation
source. Per default it is NOT changed, so you are trying to update from
the iso, which doesn't work.

Try setting
	tlmgr option location http://mirror.ctan.org/systems/texlive/tlnet/tldev
that will change the installation source (the default one) to the above
path. MIND that this will change with the final release.

After that you can call
	tlmgr update --all

Or, if you do NOT want to change the default installation source, you
can select it for one command only by
	tlmgr -location http://mirror.ctan.org/systems/texlive/tlnet/tldev update --all

All this can easily access by
	tlmgr --help
